Surface properties of Sendai virus envelope membrane have been measured, using both biological and biophysical techniques. Both normal and trypsin-treated virus were studied. SDS gel electrophoresis showed cleavage of the F protein exclusively by trypsin. The major activity change was observed in the hemolysing activity which is an expression of F protein. Hemolysis was reduced to less than 10% of its value for intact virus. 31P nuclear magnetic resonance studies of the envelope surface of the native virus showed a highly restricted phospholipid headgroup environment. Interestingly, this restriction was relieved by treatment with trypsin. Thus these data suggest a role of the F protein of Sendai virus in tightly organizing the surface of the viral envelope membrane. 相似文献

生境质量是反映生物多样性状况与局地生态功能的重要指标,在高质量发展背景下研究区域生境质量的时空演变具有重要意义。以浙江省为研究区,基于InVEST模型、热点分析及地理探测器模型探究生境质量的时空演变与影响因素,并利用生境质量结果对浙江省生态红线开展了定量评估。结果表明:(1)2000-2015年,浙江省生境质量均值呈减速下降趋势,空间上形成了西北、西南、中东高和东北、中部低的分布格局;生境退化度呈现"中心-外围"的圈层辐射结构。(2)热点分析显示,生境质量与生境退化度在乡镇尺度上集聚特征相似、冷热点空间分布趋势相反。(3)地理探测分析发现,地形(高程、坡度)是影响浙江省生境质量的主要因素,植被因素(NPP、NDVI)的贡献度随时间推移逐渐增大;浙江省生境质量空间分异受到自然因子与社会经济因子的协同作用。(4)浙江省生态红线的生境质量整体较高且稳定,不同红线类型的生境质量存在差异;高生境质量区与生态红线的错位区域主要分布在浙西南、西北部山区,而北部、中部以及东部相对较少。基于此,对生态红线调整、区域生态功能区划提出对应的策略,以期提升浙江省生态空间管控。 相似文献

Anionic vesicles containing acidic phospholipids are known complement activators. To clarify which negative physicochemical electrostatic charges on vesicles and structural specificities of acidic lipids are critical to complement activation, the electrostatic properties and activity to complement of two anionic vesicles modified with a carboxylic acid derivative or a conventional acidic phospholipid were compared. Electrophoretic mobility measurements indicated that the negative zeta potential and the electrostatic interactivity of these two anionic vesicles were equal at pH 7.4. However, the infusion of vesicles containing acidic phospholipid induced significant complement activation, while vesicles containing the carboxylic acid derivative failed to activate complement. These results indicate that the negative charge on the surface of vesicles is not critical for the activation complement, suggesting that complement activation is specific to the structure of acidic groups. This finding is likely to be important to the design of anionic biointerfaces and may support the promising medical applications of this anionic vesicle modified with a carboxylic acid derivative. 相似文献

Microbial phytase is used to reduce the environmental loading of phosphorus from animal production facilities. The limiting
factors in the use of this enzyme in animal feeds can be overcome by solid-state fermentation (SSF), which is a promising
technology for commercial enzyme production with lower production costs. Inoculum quality and the influence of inoculum quality
on phytase production are important factors which need in-depth investigation before scaling-up of high-yielding fermentation
process. A full factorial experimental design for 240 h with sampling at every 24 h was used to determine the effects of the
treatments, inoculum age (plate and liquid culture), media composition and the duration of SSF on the production of fungal
biomass and phytase in SSF systems using Aspergillus niger. The optimal treatment combination for maximal phytase production was determined by statistically comparing all treatments
at each sampling time. Both 7- and 14-day plate cultures and M1+ medium composition with 72-h-old liquid inoculum treatments
resulted in optimal phytase production at 144 h of SSF, which was the shortest duration observed for maximal phytase production.
This resulted in maximal phytase production with a mean of 884±121 U/g substrate, while the maximal phytase production observed
at 216 h of SSF (mean phytase activity of 1008±121 U/g substrate), with the same treatment combinations, was not statistically
significant from that at 144 h of SSF. Phytase production was strongly growth-associated with younger inocula. The significant
treatment variables, age of liquid inoculum and the duration of SSF, were used to predict the system response for phytase
production using response surface methodology. From the response surface model, the optimal response of the experiment was

Temperature affects both the biology and morphology of mosquito vectors. Geometric morphometrics is a useful new tool for capturing and analyzing differences in shape and size in many morphological parameters, including wings. We have used this technique for capturing the differences in the wings of the malaria vector Anopheles superpictus, using cohorts reared at six different constant temperatures (15°, 20°, 25°, 27°, 30°, and 35° C) and also searched for potential correlations with the life tables of the species. We studied wing shape in both male and female adults, using 22 landmarks on the wing in relation to ecological parameters, including the development rate. The ecological zero was calculated as 9.93° C and the thermal constant as 296.34 day‐degrees. The rearing temperature affects egg, larval, and pupal development and also the total time from egg to adult. As rearing temperatures increased, longevity decreased in both sexes. In An. superpictus, Ro value and productivity correlated with the statistically significant gradual deformations in the wing shape related to size in both sexes. These deformations directly linked to differences in immature rearing temperatures. Analysis using PCA and UPGMA phenograms showed that although wings of females became narrower dorsoventrally as the temperature increased, they became broader in males. Comparisons of the wing landmarks indicated the medial part of the wing was most affected by larval rearing temperatures, showing relatively more deformations. Algorithmic values of the life tables were determined in correlation with the results of geometric morphometrics. Identifying protein–protein and other proximal interactions is central to dissecting signaling and regulatory processes in cells. BioID is a proximity-dependent biotinylation method that uses an “abortive” biotin ligase to detect proximal interactions in cells in a highly reproducible manner. Recent advancements in proximity-dependent biotinylation tools have improved efficiency and timing of labeling, allowing for measurement of interactions on a cellular timescale. However, issues of size, stability, and background labeling of these constructs persist. Here we modified the structure of BioID2, derived from Aquifex aeolicus BirA, to create a smaller, highly active, biotin ligase that we named MicroID2. Truncation of the C terrminus of BioID2 and addition of mutations to alleviate blockage of biotin/ATP binding at the active site of BioID2 resulted in a smaller and highly active construct with lower background labeling. Several additional point mutations improved the function of our modified MicroID2 construct compared with BioID2 and other biotin ligases, including TurboID and miniTurbo. MicroID2 is the smallest biotin ligase reported so far (180 amino acids [AAs] for MicroID2 versus 257 AAs for miniTurbo and 338 AAs for TurboID), yet it demonstrates only slightly less labeling activity than TurboID and outperforms miniTurbo. MicroID2 also had lower background labeling than TurboID. For experiments where precise temporal control of labeling is essential, we in addition developed a MicroID2 mutant, termed lbMicroID2 (low background MicroID2), that has lower labeling efficiency but significantly reduced biotin scavenging compared with BioID2. Finally, we demonstrate utility of MicroID2 in mass spectrometry experiments by localizing MicroID2 constructs to subcellular organelles and measuring proximal interactions. 相似文献
